Anyone heard of Clomipramine , please!

Sorry to be a pain and post again, but just in case no-one noticed my first note, is there anyone out there who can tell me about Clomipramine? I've been put on it for the first time. And after my first dose, I was very, very drowsy for the next day. Is this normal?

Hi Lisa, how rae you doing? I've read your thread in the past titled Elephant and I felt for you.
I'm in a big battle with the weight gain myself.

Anyway...clomipramine(Anafranil) is a tricyclic with both antidepressant and antiobsessional properties. It acts on norepinephrine and serotonin systems, and may have sedative effect.
I've tried it in the past, and I hated because of the sedation.
The alternatives to Anafranil are Prozac, Zoloft and Luvox.
Stay away from Paxil, as it causes weight gain.

Google returns 10,423 hits on clomipramine,
I noticed several were user reports.

Thank you very much Anna and James for replying. Your information was very useful, Anna. So it sounds as if it's not unusual for me to be feeling drowsy with this medication. I visited the States the Christmas before last, and had a terrifying exprience where I nearly died. I'm having to go to the States again this Christmas, and I am very scared. I guess he's giving me this stuff to build up a defence for when I go. Hope it works!

Lisa,

I've been on and off clomipramine for the past ten years. I really rate it as an AD; it's certainly worked for me. In my experience, the level of debilitating sedation is dose related and does decrease markedly at any dose after a couple of months.

Lisa - Clomipramine (Anafranil™) is a tricyclic antidepressant (TCA). All TCAs bind to the serotonin reuptake receptor (like SSRIs, eg Prozac™) and the norepinephrine reuptake receptor (like reboxetine), but their affinities (ability) to bind to these receptors differs. Clomipramine is the TCA that most selectively binds to the serotonin reuptake receptor, very similar to the SSRIs.

In regard to the SSRIs, I guess you could say that clomipramine closely resembles Paxil™ (paroxetine). Clomipramine does cause more sedation (especially for the first 2 or 3 weeks) and can cause heart problems in overdose. Clomipramine works great in OCD and I have seen some success in panic disorder, as well as being a decent antidepressant.

Thank you, Cam, for your reply. Do you think that if I take this stuff from now until I go to the States in December that I will have built up some sort of protection so I won't have panic attacks when I get there? Or should I start taking it nearer the time of my trip? Is there anything else you can recommend to keep me calm? I've just starting seeing a psychotherapist, and he tells me that I'm suffering from Post Traumatic Stress Disorder...

Hi Cam.

Clomipramine (Anafranil) is a potent SRI but is not selective. It also potently inhibits the reuptake of norepinephrine, as do most of the other tricyclics (trimipramine is the only one I know of that does not). At one time, it was considered to be the most effective marketed antidepressant in the world.

Clorgyline, a reversible selective inhibitor of MAO-A, is the most effective antidepressant in the world. Unfortunately (DAMN IT), it was never marketed and probably won't be. Any ideas on how we can pursuade a drug company to persue it? I don't believe it is patentable.

Both drugs block it much less than they block the serotonin reuptake receptor. Paxil has been shown to block the norepinephrine reuptake receptor to a signicant degree in vivo, but still less than clomipramine.

Lisa - I would start taking clomipramine soon. Sometimes it takes longer for it to work in PTSD. Also, you will want to get the start-up side effects out of the way (eg anxiety, dizziness, dry mouth, headache, upset tummy, etc. - all usually mild and resolve on their own). This will take a couple of weeks, as well. Talk with your doc about this and get his/her opinion.

What are the absolute binding affinities to the norepinephrine transporter of clomipramine, imipramine, desipramine, and amitriptyline?
