I was on clomipramine - 150mg for 10 years in the 90's. It worked graet but my p/doc switched to see if some side-effect would disappear which they did.
With my current cocktail, zoloft 50mg, lamictal 100mg and zyprexa 5mg, I feel like I did when I was on the clomipramine...less the side-effects?
Any of you chemical experts have an opinion as to why? Would my current combination hit the neros in a similar fashion?

Hi Rick.I truly am no expert. You would know that if you were to read just a handful of abstracts coming from the NLM Medline (Pubmed).
Anyway, one thing that the treatments have in common is that both clomipramine and Zoloft are potent reuptake inhibitors of serotonin (5-HT). As for the other two drugs, I really can't say how they might help, if at all. For my bipolar depression, Lamictal does help partially and consistenly. It is often used by others as an antidepressant. Zyprexa helped me partially and transiently. It is often used in combination with Prozac, an SSRI like Zoloft to produce an antidepressant response. So there is undoubtedly an antidepressant potential to all three drugs that you are taking.
The two things lacking in you new treatment regime that you had in your old one is norepinephrine (NE) reuptake inhibition and anticholinergic effects. This is probably why you are experiencing fewer side effects.

Also, I thought Zyprexa worked with (NE) reuptake inhibition? I feel like my motivation/energy level has increased so maybe its the Lamictal? Told you we count on you!!

Hi Ricker,
Maybe it feels like clomipramine solely because you are no longer as depressed.
Just out of interest how do you feel without the Zyprexa and just with the lamictal and Zooloft?

> DeniseHi Denise, without the zyprexa, I get very agitated! And, without the lamictal while taking just zyprexa/zoloft, I feel flat, lethargic, so the two (zyprexa/lamictal) seem to counter each others nagative?

The combination of zyprexa and zoloft will in many ways be similar (on paper) to anafranil. As SLS mentioned, they are both potent SSRI's. Clomipramine is also a 5-ht2a/c receptor antagonist as is zyprexa.
Also, clompiramine actually has some mild/moderate d2 receptor antagonism (which makes it a mild antipsychotic). A similar (but perhaps more potent) effect will be replicated with the zyprexa.

Although I wouldn't personally worry about genotoxicity in real life, clomipramine is one of the series of TCAs that has shown such in fruit flies.

I personally think the genotoxicity thing is worth being concerned about. I think it has been replicated in more than just fruit flies.

I remember reading that the genotoxic TCA's had genotixc effects on rat bone marrow tests. You don't shrug off carcinogens IMHO. Sure everything is fine and dandy till you end up with tumors 20 years down the road.
