Hi all, I've been watching the development of this drug like a hawk since I first found out about it around a year ago. Its been under re-examination by the EMEA for ~1 year now and rumour has it that they are very close to approving it. See http://en.wikipedia.org/wiki/Agomelatine for a rough overview.
I'm starting this topic to see if anyone out there has any new information on Agomelatine. I would really like to hear from you if you have tried the drug in a clinical trial.
What was your opinion of the drug for depression or anxiety? How does it compare to current SSRI or Tricyclic antidepressants? Where there any side effects to speak of? Im sure a lot of people really want to know if there are any sexual side effects with it (it has none apparently)?
I would also like to hear from you if you have any information on how close this drug is approval in EU or USA.Hope to hear from some of you soon!

I don't know more than you but can I ask you why you're more interested in Agomelatine than in order drugs in the pipelines ?Here's a list just in case you're interested.

Hi Marty thanks for the list, yeah I often check that website thanks. I'm interested in Agomelatine for a couple of reasons. Firstly it is allegedly as effective as current first line treatment (SSRIs) but has a much much better tolerability profile. I've been on SSRIs for GAD for about 5 years now and I'm fed up with the side effects , so this sounds very attractive to me. Secondly out of the other drugs in development right now Agomelatine is the closest to market here inthe UK. Although I have to admit Saredutant and Amibegron sound promising as well, but they are a couple of years away(if not more), especially since they are not submitted to the EMEA yet, only the FDA.

This is also why I'm interested into it. I can't stand the side effects comming from typical serotonergic antidepressant. They only thing that makes be ambivalent toward Agomelatine is that Melatonin makes me depressed in a couple days and I wonder if it is significant or not. I'm not sure it is and to bring a comparison that sum up my logic: Tryptophan(5-HT precusor) makes me anxious but SSRI is anxiolytic to me. Stimulating a receptor by ingesting his precursor (ie Tryptophan) or his own endogenous neurotransmitter (Melatonin pass the BBB, Serotonin does very poorly) does induce the same effects as inhibiting his reuptake .. and so I may as well forget my reaction with Melatonin and stay positive toward Agomelatine.... IF you have some thoughts about it plz share.Btw, if I ever make some "Side Effects Suck!" T-Shirts.... want one ? ;)

Hi Marty....off the top of my head.....
I havent tried melatonin, as whenever I couldnt sleep on SSRIs Id either reach for Nytol, Zoplicon or "insert downer here". As a result Ive never really paid it that much attention untill now ive started reading about Agomelatine. I suspect that Melatonin has secondary action on other neurotransmiter recptors that could account for the depression you spoke of.
As for the anxiety you experienced from ingesting precursors like trytophan. I would put this down to the unselective action of natural serotonin. Ie your increasing overall levels of serotonin in all synapses and all types of serotonin receptors at once. Certain types of 5HT receptors are know to increase anxiety when stimulated. Indeed, this is why many ppl experience increased anxiety when first starting on an SSRI, although this reduces after about 2 weeks when the recptors desensitise to the increased serotonin in the synapse. Another thing to note is that selective serotonin reuptake inhibitors are called so because they are selective and only target a few types of 5HT recptors. This could account for improvement over precursors and older ' less selective' drugs.
So yeah now we have Agomelatine, something totally new. This drug primary acts on the M1 and M2 receptors as an agonist, helping you reset your body clock and get some sleep. I personally am not really interested in this aspect of the drug....yeah ok its cool if I sleep well but its not so much of a problem for me. Im much more interested in the drugs secondary acion as a 5-HT2C antagonist, and 5-HT2B antagonist. Its this action which indirectly accounts for the drugs alledged antidepressant/anxiolytic effects minus all the regular side effects of antidepressants to date....Brilliant! I just hope it lives up to the hype. Of the 2 reports Ive read online both were very positive, and I cant wait to give it a go. :)ps I wouldnt worry this aint no melatonin. :)

Yes. There's more to M1 and M2 than sleep regulation / Circardian regulation.

Indeed, it could be just that. Serotonin, just like dopamine, has a dichotomous nature. At least at the receptor level and their induce effects; "Serotonin = feel good" is oversimplistic and is just bad pop-science promoted by years of big pharma ADs marketting.

I have to disagree.
Quoting Wikipedia:"SSRIs are described as 'selective' because they affect only the reuptake pumps responsible for serotonin, as opposed to earlier antidepressants, which affect other monoamine neurotransmitters as well. Because of this, SSRIs lack some of the side effects of the more general drugs."
SSRIs target every 5-HT receptors to some extent.

Indeed. Other receptors affinity (ie Histamine) account for TCA and other less selective drugs harsh side effects.

And some antipsychotics also. "for the drugS" ? you mean SRIs or Agomelatine ? for Agomelatine it's fact, but for other SRIs most doesn't antagonize 5-HT2A/5-HT2B except Serzone and somehow Tianeptine which by accelerating reuptake of 5-HT reduce 5-HT2A/5-HT2B (etc) stimulation.I wonder if upregulation doesn't make those kind of ADs even more suceptible to poop-out.

Marty, Interesting about the Melatonin. I didnt realise it was being tested with depression. Still even if it does have some success with depression it probably wont to jack for anxiety (which is my main concern, ive got GAD). I also wasnt aware that SSRI targetted all 5HT receptors to some extent. That just makes me think that Agomelatine sounds even better because its not a 5HT like molecule it should only target the recptors it supposed to, limiting the side effects.
You asked if Agomelatines been pushed as a sedative. Well they claim that it will sort out your circadian rhythm whithout causing any increase in day time drowsniness....Bonus!
By "for the drugs" I did indeed mean agomelatine, currently not other drug out there targets this particular 5HT receptor combination, so its all new teritory. As for up regulation and poop out, this isnt something I know very much about....Ive not experienced it yet either (thankfully). I read somewhere once that SSRIs can fail on you after about a decade. Ive had to recently switch from citalopram to a TCA because it was giving me gastritis. So that sucks, TCA isnt as effective and i get drowsiness.

The other one I saw was a comment at the bottom of a article on agomelatine. I cant for the life of me find it again. But she was claiming to have been on it for a year in a clincal trial and said it was a completely "Blissful year", she was worried about haivng to change back to a SSRI when the trial finished.Now Im just hoping we get some more information on here about it. theres gotta be plenty of ppl out there , come on tell us what you think :)

Thanks Neg! ... I've post on that thread. I really hope the FDA doesn't screw this one up :
"Agomelatine was discovered and developed by the European pharmaceutical company Servier Laboratories Ltd." ... European company and the product has no side effect while being as effective as Paroxetine ? The FDA must know that this while cost US big pharma BILLIONS pretty quickly... that is scary and I think that not enough spotlight is on that drug in the news/magazine etc .. being more 'anticipated' would put -a little- more pressure on the FDA.
If that med doesn't get approval here but gets it in Europe I wish we would be legion to fight the case in America.
