Zuranolone is a positive allosteric modulator of the GABA-A receptor. I guess it makes the receptor more sensitive and more apt to respond when GABA is received. It is related to brexanolone, the drug infused i.v. to treat post-partum depression. Zuranolone is taken orally.
The early studies of zuranolone reported positive results in treating Major Depressive Disorder and Bipolar Depression in addition to post-partum depression. However, a more recent trial failed to show efficacy for MDD or BD. After the announcement of the study, Sage Pharmaceuticals' stock plummeted on the stock market.
The thing is, the trial lasted for only 15 days. What the hell? I don't know whether or not Sage stopped development, but given the latency of response seen in almost all antidepressants, a 15-day trial is incredibly stupid and criminally irresponsible. If zuranolone is proven to be genuinely useless, it won't be because of the results of this trial. The only thing genuinely useless designing a clinical trial that lasts only 15 days.
Anyway, does anyone know what the drug company plans to do with zuranolone?

It looks like Sage hasn't given up on zuranolone for major depression.

Thanks, Hugh!
I hope you are doing well. I started taking Marplan two days ago. I hope I can get my doctor to be aggressive with dosage. The last time he worked with it, I think the recommended maximum dosage was 30 mg/day.

You're welcome, Scott. Good luck with Marplan.
I've been feeling better than usual lately. Do you remember posting this?

There's a new supplement derived from fish oil called specialized pro-resolving mediators (SPMs). I really like it and I think you should give it a try. It's head and shoulders above any fish oil supplement I've tried before, and I've tried many. Here's an article about it:
