Every time I read about a new AD being tested on
clinicaltrials.gov I just shake my head. I can't get excite about a new reuptake inhibitor anymore. The newest antidepressant Viibryd just make me think "meh". But here's a really neat one that's in the works, I think it could be the new prozac. Something that could revolutionize AD medication treatment.

I sometimes think that a smaller, "shriveled" hippocampus is a true atrophy of tissue that occurs for lack of use. This lack of use is the result of global reductions in cerebral brain activity. In other words, a smaller hippocampus is the result of depression rather than its cause. Perhaps there are reciprocal pathological processes that evolve whereby hippocampal atrophy acts to reinforce the depressive state, which then allows for greater hippocampal atrophy.

Just a thought.

Since the MRI's of the brain I've had show normal for age I guess it hasn't shunk? Phillipa

Aren't stress hormones (especially cortisol) thought to be the cause of damaged, smaller hippocampuses?

I was thinking the same possibility. Or perhaps something such as chronic stress or some other factor affects (and shrinks) the hippocampus, which starts off or contributes to the start of depression. Maybe the cause and effect arrow is different for different people. But this drug might have some real utility in either case.

Sometimes I get a headache. I know something is causing it, but I go ahead and take ibuprofen. Usually when the ibuprofen wears off, the headache doesn't come back. Either the ibuprofen somehow also gets at the cause of the headache, or just relieving the pain allows the body to recover from the thing that's causing it. For depression, maybe this drug acting by improving hippocampal volume can help kick start a bunch of healing processes.

I'm just glad someone is trying to come up with something new.

That's a good alternate explanation. Isn't that the generally advocated theory?

I wonder if this NSI-189 targets the neuritin receptor.

* I just performed a Google search. The drug doesn't seem to target that site. Neuritin would make an interesting peptide to study, though.

Yeah. I would love to jetison the 7 drugs I have to take for one that uses a more direct mechanism.

Judging by where its at in clinical trials, any thoughts on when it might up for possible approval and then prescribed?
