Hi there, I was wondering if anybody here is in a clinical trial for lundbeck's new drug 'Lu AA21004'?
It's a 5-HT3 antagonist and 5-HT1A partial agonist and currently in phase III in the USA.
http://www.takeda.com/press/article_28813.html
Theres not much recent news on the drug since 2007, but there are plenty of clinical trials underway on :
http://clinicaltrials.gov/ct2/results?term=Lu+AA21004It sounds very promising for depression and axiety.
So is there any one out there taking it? How effective is it, and how are the side effects etc?
Hope to hear from you

I've not heard of this med but not surprising. No I'm not in a trial how did you hear of this med? Interesting no name really assigned yet either. Phillipa

i prefer the sound of this drug over that other 5-HT1a agonist in the pipeline, vilazodone i think it is called, which seems to lack the effects of a real 5-HT1a agonist in vivo. it's actual relevant action may be it's supposed "secondary" SRI effect. just what we need, another SSRI to waste time promoting.
this drug, Lu AA21004, appears to be a real 5-HT1a agonist, that is to say it releases catecholamines in the PFC as associated with the actions of an 5-HT1a agonist. curiously however, it also increases extracellular serotonin, an effect very unlike what is expected with 5-HT1a agonism, which is supposed to decrease extracellular serotonin concentrations. this may be an effect of the 5-HT3 antagonism, i would like to know more about that, and where exactly the extracellular serotonin concentrations would be increased and what the neuropsychiatic complications may be understood or theorised as being.

Yeah Id prefer a new mechanism of action over a SSRI any day. PArticularly since 5-HT1a agonism is particularly good for anxiety (I have GAD) Although vilazodone is an SSRI with no reported sexual side effects (yet)....maybe not so bad? Ill probably try both anyway :)

Wouldn't this drug just be like combining Remeron with Buspar?

No remeron acts on several other 5HT receptors causing large weight gain and severe daytime drowsyness as side effects (at least it did when i took it). Buspar isnt a very good 5-HT1A agonist, which is why its not very popular.
Lu AA21004, was gettin good results in phase 1 and 2. Its something a bit different.
Not much information on side effects yet though. But we can only assume they will be hoping for no sexual dysfunction and negliable weight gain like most of the new drugs in development.> Wouldn't this drug just be like combining Remeron with Buspar?

Hi
Ok, thanks for Clarifying that for me. Remeron never actually caused weight gain or excessive daytime sleepiness for me so I'm not that optimistic about a similiar drug with or without the side affects.Lets hope it is a stronger antidepressant than either Remeron or Buspar.
