We might finally get a 5-HT1a partial agonist that doesn't block dopamine receptors. Vilazodone also acts as a SSRI.
I'll be curious to see if vilazodone also proves useful in treating anxiety disorders.

Hmm... sounds interesting. Does it work similarly to Trazodone and Serzone? I know they have a dual serotonergic action as well, just can't remember what exactly it is. Do you know when it's expected to be released?

Actually, the drug is similar to gepirone, an investigative drug that has failed to be approved by the FDA. I believe that the reason buspirone can act as such a good augmenter of antidepressanats is because of its ability to act as a partial agonist (stimulator) of 5-HT1a receptors.
The article said that the drug company will submit vilazodone for approval later this year.

Scott are you considering trying it or just educating? It's said SSRI's help anxiety but they never have with me would it be similar to that? And how are you doing? Love Phillipa

I am very much looking forward to vilazodone. I wish they would have approved gepirone, as I think it would have been a great add-on med. It doesn't appear to have sexual side-effects - this would be a godsend for many of us. Fingers crossed!

it seems that it may only have 5-ht1a effects in vitro. its lack of effect on extracellular dopamine and noradrenaline is not shared by 5-ht1a partial agonist monotherapy, or when augmenting an ssri.

Which 5-HT1a partial agonists DO increase extracellular catecholamine concentrations via that mechanism?

Wow. That's some pretty big news.As was said, it is too bad that gepirone was shot down by the FDA. There is a drug called flesinoxan that is a partial agonist at 5-HT1a receptors. I like this drug because, unlike gepirone, it is not metabolized into a NE alpha-2 receptor antagonist. Two such antagonists made my depression worse.

ALthough Pubmed is very reliable, there are so many contradicting articles on there. This thread got me looking at the articles I have bookmarked, and I think this may be the one of greatest importance.

Note Vilazodone is NOT the same as Valdoxan (or Agomelatine, the one that works on melatonin). Apparently Vilazodone had very high HAMD scores in trials and very little sexual disruptions.
