New treatment for ADHD: Connexyn

Licensing application submitted to FDA for extended release guanfacine (Connexyn) for ADHD

Shire plc has submitted a new drug application to the FDA for its investigational drug guanfacine extended release (SPD503, Connexyn™), which, if approved, would be the first once-daily selective alpha-2A-adrenoceptor agonist for the treatment of ADHD in children aged 6 to 17 years. Guanfacine is not a CNS stimulant or a controlled substance and is claimed to have no known potential for abuse or dependence.

Hi ed,
Thanks for posting that. Can you say more about how this might help those with ADHD? I'm a bit clueless about the receptors and such beyond a basic knowledge.

My ADHD symptoms have been worsening lately, and I'd like to have an option beyond Provigil as long as I'm also on Nardil (which is also something I'm reconsidering).

Although looking at Epocrates on my palm, I see that use with MAOI's is contraindicated for guanfacine, so I would expect that this new drug would also be contraindicated. (sigh)

Crazy how you will get drugs like clonidine and connexyn which help ADHD in that they lower noradrenaline, wheras you get straterra which raises it, and both are effective for ADHD.

Is it correct that guanfacine mainly treats hyperactivity, and not so much inattention? That's the impression I've gotten.

So what if you have innattention and innactivity, and you don't live in the USA?

You could try having guanfacine prescribed off-label. In the US the brand-name is Tenex, used for high blood pressure. I'd imagine it's widely available for high blood pressure.

BTW my impression is it's not very effective for inattention, but I could be mistaken.

there are articles on it improving cognition i am very sure. dr. jay a. goldstein, if i remember correctly, also seemed to like the drug.

seems like it may be also effective for anger, but i am not sure, maybe it isn't like clonidine that way.

i suppose one might hope it has no sexual side effects, as it may, but i am not completely sure.

that this drug had better come out in canada. grr.

worsening or causing depressive symptoms may also be a side effect, but i don't really care. i want a brain.
