If you have no bipolarity, I would start at 300 mg/day. Kidney and thyroid adverse effects are both dosage-dependent. At 300 mg/day, the risk is close to zero.
300 mg/day is magic for me in treating my bipolar depression. However, low-dosage studies have shown that it is equally effective for unipolar depression. When I increase the dosage to 450 mg/day, everything falls apart, and I relapse. I believe this bimodal dossage-response curve involves glutamate.
I strongly recommend that you start at 300 mg/day for two weeks. If nothing happens, 450 mg/day would be your next move.
