Medication worked at one point... now it doesnt

Has this ever happened to anyone? You take a medication for years successfully, then have to go off of it for one reason or another (for example side effect or new med in your cocktail)... then you try to go on it a few years later, and the med doesn't work!? That really stinks... just my two cents... I suppose it can be a number of reasons... life circumstances, age, other medicines, etc..
