In reply to Prazosin causing/worsening depression?, posted by uncouth on April 10, 2012, at 14:52:29
> I've been on prazon for the last i think 5 or 6 days, trying it for treatment refractory insomnia. I take 6-10mg at night. Since sunday, I have experienced a severe worsening of my depression, feeling worse than I have in months and taken to bed. Sleep has not responded to prazosin and i still require ambien cr, and if that doesn't work, then seroquel.

> But i'm wondering if the prazosin at night would in any way cause or worsen depression during the day?I don't know the answer to your question for sure, but the use of prazosin in treating depression is uncommon. Just like with other neurobiological treatments, almost anything is possible given the unique differences between the biologies of individuals. What you are doing is performing an experiment. It is important to take note of your experiences as they occur in time and in association with treatment changes. Prazosin might be good for me, but not for you. Clonidine reduces NE activity, and is well known to cause depression. Prazosin reduces NE activity also, albeit by a completely different mechanism. I believe Phiddipus can elaborate on this regarding how these drugs differ in their effects on the prefrontal cortex (PFC), a region that is important in the cascade of events relating to depression and its treatment. Personally, it is my feeling that prazosin reduces the overactivity present in the subgenual cingulate that has been observed in depression. With catecholamines like dopamine and norepinephrine, sometimes less is more.
I found prazosin to produce fatigue and tiredness at first. I was actually fooled into thinking that this was a worsening of depression. It took 7 - 10 days for these things to dissipate, but they completely disappeared. Perhaps your experience will be similar.
