C40 et al,

Yes, you have it right, C40. I take 7.5 mg/day of Deplin with my other meds. Deplin provides a small positive effect on my mood, but nothing like it did at start-up. However, if I take a break from Deplin for about a week, then I experience a very pleasant, peaceful mood when I restart Deplin. This restart response is similar to my original start-up response, just not nearly as profound.

The main reason that I continue to take Deplin is to offset the folate loss due to taking antiepileptic medications. And, secondarily, I think it provides me some amount of an antidepressant effect.

ATTENTION ALL DEPLIN USERS: It is VERY important to take methyl-B12 with your Deplin. In the absence of B12 supplementation, folate (in this case L-methylfolate) depletes the body of B12. The very best form of B12 to take is sublingual methyl-B12 (methylcobalamin).

Also, P-5-P (the coenzymatic form of B6) should be taken with Deplin.

There is good reason why Metanx contains both methylcobalamin and P-5-P to go along with the L-methylfolate.

Methyl-B12 is the coenzyme that interacts with L-methylfolate and feeds the Methylation Cycle. So, if we continue to take Deplin over an extended period of time without also taking methyl-B12, then B12 will become in short supply. Without sufficient B12, the L-methylfolate gets stuck in the methyl trap. Further, all kinds of serious problems occur due to B12 deficiency.
