The point of the trial is to see if the study drug is effective -- which means you have to test it alone. It doesn't make a huge amount of sense, from the drug company's standpoint, to run tests to see if their drug is only effective if combined with other drugs -- they have to prove that the drug is valuable on its own.
Thus, you have to stop all other drugs in order to be included in the study.
Does that make more sense?
