My doctor has used Strattera, another NRI, extensively, and related to me at my last appointment that his observation is that it does not seem to produce a robust antidepressant response. I wonder if some amount of serotonin reuptake blockade is required in combination with the NE reuptake blockade. Desipramine is selective for NE reuptake but also has a relatively low dissociation constant for blockading serotonin reuptake in human cells, between 10 and 100, I believe.
