Hi!
After a single dose, diazepam has a short duration of action because it is rapidly redistributed into fat, removing it from the blood. After repeated doses, diazepam has a long duration of action, it's long-acting active metabolites accumulate and an equilibrium is achived between fat deposits and the blood.
