The downregulation does not mean it wears off. Infact the alteration in receptors can occur when the drug works.


For instance. SSRI's downregulate the 5-ht1a receptors, but this is one of the things that is believed to be associated with the responce. Ie post mortem brains have shown to have significantly upregulated 5-ht1a receptors.
