I need to take 300mg of imipramine or desipramine in order to benefit from it. I find that if I miss a dose, I begin to relapse within 12 hours. Also, I recapture the improvement within 15-30 minutes after taking an adequate dose. To answer your question, I think it is quite possible that you are gaining and then losing an improvement that desipramine might be providing you. I think it makes sense to push the dosage.
