Abstract
Emerging evidence indicates that NLRP3 inflammasome-induced inflammation plays a crucial role in the pathogenesis of depression. Thus, inhibition of NLRP3 inflammasome activation may offer a therapeutic benefit in the treatment of depression. Metformin has been shown to have potential anti-inflammatory activity, but the underlying mechanisms remain obscure. We used a chronic mild stress model of depression and cultured primary macrophage to investigate the effects of metformin on depression and its underlying mechanisms. We demonstrated that metformin alleviated depressive-like behaviors in the chronic mild stress-induced anhedonia model of depression. We further found that metformin significantly suppressed NLRP3 inflammasome activation, subsequent caspase-1 cleavage, and interleukin-1β secretion in both peripheral macrophages and central hippocampus. Our findings reveal that metformin confers an antidepressant effect partly through inhibition of peripheral and central NLRP3 inflammasome activation. In light of metformin favorable properties, it should be evaluated in the treatment of depression and related neurologic disorders characterized by NLRP3 inflammasome activation.

I took Metformin for 2yrs while on Nardil (to prevent wt gain). I wouldn't say my mood was great during that period so for me I don't think it was very effective as an AD.That said, it did help keep Nardil wt neutral, and maybe it was doing more than I realized. I'm restarting a low/mid-range dose today (500mg and then up to 1000 in a week or so). It protects the heart and lungs which imho is helpful should I come down w/ COVID (please god no). One retrospective study of Metformin found reduced diabetics who were taking metformin vs. those who were not.
Either way, it's helpful for preventing med induced weight gain. A lot of pdocs won't prescribe it since they consider it a diabetes medicine. If you need a study to print and show your doc that it may have beneficial mental health properties as well, perhaps this is one.

Most depression studies in mice are a 'stress induced' depression model. Metformin appears to help in this model of depression, but in humans, there are likely many other causes.

I basically turned the course of "illness" around after Metformin was added. However it is hard to say why it did. Could be the Metformin, could be the awesome rehabilitation facility. I stayed there for a year. It is often good for weight gain and it certainly does no harm. Very few side effects. Have to get some kidney blood work every once in a while and that is about it. Kidneys should be healthy.
