Lumateperone affects multiple systems (serotonergic, dopaminergic and glutamatergic) in the central nervous system. It acts as a antagonist of 5HT2A 0.54nM receptor and modulates D1 4.1nM and D2 3.2nM dopamine receptors where it is a partial agonist at presynaptic and antagonist at postsynaptic receptors. Serotonin transporters at 3.3nM. Through the mTOR pathway, lumateperone augments both NMDA and AMPA activity. Because of its complex pharmacology, it is not clear which of these activities are primarily responsible for its antidepressive and antipsychotic activities.

Very interesting.

I wonder if I would get serotonin Syndrome. I am on Venlafaxine 37.5 and Trazodone 75. If it wasn't for that fact, I'd try it in a heartbeat. No weight gain, right? But it could have sexual sides because it is a Serotonin Reuptake Inhibitor.

Now that I have started studying, time goes by much faster. I am not lingering around anymore waiting for better drugs to come out. They just come out...

And I do think that this decade, there will be something helpful for me. Aloradine for social anxiety! Please, PLEASE be good! It's not that far away from coming out I think!

I avoid many social situations... I'm not trying to become a politician or anything but I sure would like to go out into the world and connect with people to some degree. Living a life of solitude here...

Your dose of both these drugs is so low that I wouldn't worry the slightest about serotonin syndrome. In any case, life-threatening serotonin toxicity (ST) is exceedingly rare with only reuptake inhibitors. As a precaution, though, it may be a good idea to have some cyproheptadine (Periactin) (CHD) at hand as an antidote. Actually, now it occurs to me that trazodone blocks serotonin 5-HT2A receptors, which is the same action through which CHD works as an antidote to ST.

Magic mushroom drug could be depression breakthrough, says FDA Richard Staines.
The liberty cap mushroom, notoriously used illegally as a psychoactive drug, could be the inspiration for a new therapy for depression according to the US drug regulator. Colorado and a few other states have already decriminalized uhe use of magic mushrooms.

Mental health drug specialist COMPASS Pathways said that the FDA has designated the chemical psilocybin, which causes psychoactive effects in those eating magic mushrooms such as Psilocybe semilanceata, as a Breakthrough Therapy in treatment-resistant depression.

The FDA gives the designation to drugs where evidence demonstrates they could be a substantial improvement over available therapies for serious or life-threatening diseases.
Drugs given the designation are hastened through the development process by the FDA, which can grant a faster six-month review if it feels the evidence is still compelling once trials are completed.

The company said in a statement that the designation is a significant milestone for psilocybin therapy and psilocybin research, after many years of research by teams in the US, the UK and Switzerland.

The Heffter Research Institute was the first to fund research in this field, and supported early studies at Johns Hopkins University, New York University, and Harbor-UCLA Medical Center.

In the UK, the Medical Research Council backed the proof-of-concept study of psilocybin for treatment-resistant depression at Imperial College London.

COMPASS is now running the first large-scale psilocybin therapy clinical trial for treatment-resistant depression, which will take place in Europe and North America over the next year or so.
The company said treatment-resistant depression affects approximately 100 million people around the world.

The FDAs decision could prove a bureaucratic headache as psilocybin is deemed a highly illegal schedule 1 drug in the US, with a high potential for abuse and no currently accepted medical use but successful studies could change their approval decision

George Goldsmith, executive chairman, COMPASS Pathways, said: This is great news for patients. We are excited to be taking this work forward with our clinical trial on psilocybin therapy for treatment-resistant depression. The FDA will be working closely with us to expedite the development process and increase the chances of getting this treatment to people suffering with depression as quickly as possible.

I doubt youu would get serotonin syndrome. Those are very low doses of Effexor and Trazadone.

I will keep an eye on it. The price is probably pretty hardcore. I managed to lose considerable weight on Seroquel and Zyprexa, so if i took it, I'd take it for the effects on negative symptoms and depression. I don't know why I suddenly am able to lose weight. Will it be covered by insurance for you US guys?

Drugs, including antipsychotics, affect different people differently, and even affect the same person differently at different times. Same thing with alcohol and vitamins and supplements.

I've been able to keep my weight in check despite clozapine. I've gone up and down at times. Going down generally does require a notable but not extreme amount of determination or will power. I exploit my laziness and refrain from going to the store, where I *would* buy sweets

My doctor thinks the European Drug Administration is in no way better than the FDA. So I could get Caplyta ahead of time if the price is not a total ripoff.
