The PDR are the approved inserts by the FDA, and yes, you are right there are two ways of finding out relatively new agents. One is RelativelyNewSuperAgent.com and go and download the Acrobat PI (prescribing information) on their website. The other is combing through recent ANDA/NDA/letters of information at Drugs@FDA on the FDA web site.

For older agents though, the PDR is the only source of reliable information. RXList does have some although I notice some of the files are being converted over to a more dumbed out version.

As for PCs -- well when my doctor had a full office he had access to PDRnet, which is the subscription and what comes annually with the PDR. Also there is a smaller subset which contains just major side effects and standard prescribing amounts that can be fitted onto a PDA (don't ask me what OS, I think there were Palm ones, there may be Windows Mobile ones now.)

It has become frightfully expensive as the years have gone by but I may end up getting a new one as I haven't had a recent one in some time. Having access to an older one and a newer one can be an advantage too because some agents that have fallen out of favor may not be listed in the new ones, although I think they are mostly all inclusive except for agents that are 100% generic only, which are just listed by company and no description.
